The hottest ticket of the Festival is the star-studded Opening Night Gala, a comedy extravaganza featuring the best and brightest in local and overseas guest artists. In the Comedy Festival's 20th year, this ritzy, glitzy red carpet event takes place in the appropriately regal Her Majesty's Theatre. Hosted by Spick 'n' Specks peacekeeper and compere extraordinaire Adam Hills. Starring Charlie Pickering, Danny Bhoy (UK), Dave Hughes, Demetri Martin (US), Dylan Moran (Ire), Fiona O'Loughlin, Freestyle Love Supreme (USA), Jason Byrne (Ire), Jimeoin & Bob Franklin (The Cooking Show), Judith Lucy, Lano and Woodley, Stephen K Amos (UK), Tim Minchin, Tripod, Wil Anderson and more.

Each year as part of the Melbourne International Comedy Festival, six of finest comedians in Australia and the world go head to head in a comedy debate full of jokes, stunts and laughter. In 2006, the 17th annual Melbourne International Comedy Festival Great Debate was broadcast on Network Ten, with comedians debating the topic "Coming First Is All That Matters". Speaking for the affirmative is team captain Paul McDermott, supported by Jean Kittson and Jason Byrne. Speaking for the negative is team captain Stephen K. Amos, supported by Rove McManus and Meshel Laurie. Corinne Grant acts as adjudicator for the debate.
